摘 要:目的 寻找抑制胶质瘤细胞生长的新的相关基因。方法 应用mRNA差异显示技术与cDNAarray反向Northern杂交相结合 ,分离、克隆表皮生长因子 (EGF ,10 0ng/ml)作用后生长抑制的人脑多形性胶质母细胞瘤BT32 5细胞与对照组 (不加EGF)相比新表达或表达明显上调的基因。结果克隆了 6个BT32 5细胞生长抑制后表达明显增高的EST (expressedsequencetag) ,其中 1个为新基因序列 ,另 5个分别与CGI 5 1、转醛缩酶 (Transaldolase、TAL)、核糖蛋白L35a、 4 0s核糖相关蛋白基因及BC 2蛋白mRNA有不同程度的同源性 ,其中功能分析提示TAL表达增加可诱导细胞凋亡。 6个EST均投递GenBank ,分别获得了接受号 :AW4 0 884 4~ 4 0 884 8。结论 所分离、克隆的基因可能与胶质瘤的生长抑制有关。Objective To isolate and characterize new genes involved in glioma growth inhibition. Methods mRNA differential display and screening of cDNA expression array were performed to identify genes that are differentially expressed in BT325 cells inhibited by EGF (100ng/ml) as compared to control cultured BT325 cells.Results 6 ESTs (expressed sequence tag), which expression were highly up regulated in growth inhibiting BT325 cells, were cloned and sequenced. Homology searching revealed that one was new gene fragment and the other 5 were highly homologous to CGI 51?Transaldolase (TAL)?Ribosomal protein L35a?40s ribosomal related protein gene and BC 2 protein mRNA respectively. Of these,TAL was thought to be associated with apoptosis in recent research. Based on above findings, 6 ESTs were submitted to GenBank and received accession number from AW408844~408848. Conclusions The genes isolated and cloned may be involved in glioma growth inhibition.
