摘  要:目的 研究周围神经损伤后 ,局部应用碱性成纤维细胞生长因子 (basic fibroblast growth factor,b FGF)对脊髓前角运动神经元的保护作用。方法 切断 SD大鼠右侧坐骨神经 ,近端套接单盲硅胶管后 ,管内立即注入 2 0μl b FGF(浓度为 40 0 0 u/ ml)或等量生理盐水。术后 2周 ,观察 L 3- L 5脊髓前角外侧群大、中小型神经元乙酰胆碱酯酶 (acetylcholinesterase,ACh E)、酸性磷酸酶 (acid phosphatase,Acp)和一氧化氮合酶 (nitric oxide synthase,NOS)的阳性细胞数、反应强度和平均灰度。结果 坐骨神经损伤后 ,脊髓腰节段运动神经元因受损伤的影响发生明显退变 ,表现为 ACh E活性降低 ,Acp、 NOS活性升高。局部应用 b FGF后 ,大型运动神经元 (α运动神经元 )阳性细胞数、反应强度和平均灰度接近正常侧 ,与对照组比较具有显著性意义 (P<0 .0 1)。结论  b FGF可以有效地防止周围神经损伤引起的脊髓前角α运动神经元退变 。? Objective To study the protective effect of bFGF on spinal cord motor neurons after sciatic injury. Methods The sciatic nerve of 26 SD rats were transected unilaterally and a blind silicone tube was contacted with the proximal nerve. 20μl bFGF or saline were immediately administered into the tube. Activities and distribution of acetyl cholinesterase (AchE), acid phosphatase (AcP) and nitric oxide synthase (NOS) in anterior horn of L3 L5 spinal cord were observed 2 weeks after operation. Results Spinal motor neurons were injured after the transection, and a decrease in AchE as well as an increase in AcP and NOS activity were observed. Local application of bFGF could significantly relieve injury influence on the enzymatic activity of α motor neurons. Conclusion These results suggest that bFGF plays an important role in protecting the spinal α motor neurons against degeneration induced by peripheral never injury.\;〔
