脑膜瘤中整合素α_3mRNA表达及其生物学意义

摘　　要：背景与目的:在肺癌、黑色素瘤及脑胶质瘤等肿瘤的研究中,已证实整合素表达的改变对肿瘤的分化、增殖及侵袭性起极其重要的作用,而到目前为止国内尚未见有关脑膜瘤的整合素表达的报道。本文旨在探讨脑膜瘤中整合素α3的表达及其生物学意义。方法:用原位杂交的方法检测36例脑膜瘤组织、2例正常硬脑膜及3例蛛网膜组织中的整合素α3mRNA的表达;用SABC免疫组化方法观察脑膜瘤中Ki-67的表达。结果:整合素α3mRNA在良性脑膜瘤、不典型脑膜瘤、恶性脑膜瘤中的表达分别为2.52±0.362,1.75±0.316,1.42±0.633,不典型脑膜瘤和恶性脑膜瘤的表达明显低于良性脑膜瘤的表达(P<0.05,P<0.01),侵袭性脑膜瘤的整合素α3mRNA的表达明显低于非侵袭性脑膜瘤的表达(1.63±0.462vs2.61±0.526,P<0.01),脑膜瘤中整合素α3mRNA的表达与Ki-67标记指数相关。结论:整合素α3亚单位参与脑膜瘤的生长控制过程,随着整合素α3mRNA表达的减少,肿瘤细胞的增殖活性升高,恶性程度增加。整合素α3mRNA的表达的下调与脑膜瘤的侵袭性生物学行为有关。Background &Objective:It was reported that reported that ch anges of the expression of integrins played critical roles in differentiation,proliferation,and invassion of lung carcinoma,melanoma,and glioma.However,no data was available on the expression of integrins in meningiomas.The aim of this s tudy was to investigate the expression of integrin-α 3 in meingiomas and its biological sig nificance.Methods :Integrin-α 3 subunit was detected using in situ hybridization in meningiomas(36cases),normal dura(2cases),and arachnoid tissue(3cases).SABC immunohistchemical assay was used to determine the expressions of Ki-67nuclear antigen.Results:Expressions of integrin-α 3 mRNA in benign,atypical,and malignant meningiomas were 2.52±0.362,1.75±0.316,and 1.42±0.633,respectively.The e xpression levels of integrin-α 3 mRNA were significantly lower in the atypical and maligant meningiomas t han in the benign meningiomas(P<0.05,<0.01).The integrin-α 3 mRNA expression in those with invasive biological behavior was lower than that in those without invasion(1.63±0.462vs 2.61±0.526,P<0.01).Moreover,the expression of integrin-α 3 mRNA was inversely associated with Ki-67labeling index.Conclusions :Integrin-α 3 sub-unit participates the process o f regulating growth of meningiomas.The proliferational a ctivity and malignant grade of menin giomas correlates to decreased expr ession of integrin-α 3 subunit,and the down-regulation of integrin-α 3 mRNA is associated with the invasive biological behavior in meningiomas.
