用小鼠骨髓及胎肝微核试验评价丙泊酚细胞遗传毒性

摘  要:目的 探讨丙泊酚对细胞遗传物质可能产生的损伤作用。方法 应用小鼠骨髓微核试验及胎肝微核试验检测丙泊酚对细胞微核发生率的影响。结果 骨髓微核试验 :丙泊酚 7 5mg/kg、2 5 0mg/kg和 5 0 0mg/kg三种剂量的微核发生率分别为 (2 5 0± 1 2 2 )‰、(2 33± 1 5 1)‰和(2 6 7± 1 0 3)‰。均为阴性结果 (P =0 48)。胎肝微核试验 :三种剂量的微核率分别为 (2 39±1 2 6 )‰、(3 0 1± 1 88)‰和 (2 98± 1 87)‰ ,均为阴性结果 (P =0 2 6 )。Objective To evaluate the possible impairment of propofol on cellular germ plasm in rats.Methods The rates of micronuleus influenced by propofol were measured by mouse bone marrow and fetal liver micronuleus test.Results The rates of micronuleus in bone marrow were (2.50±1.22)‰,(2.33±1.51)‰,and(2.67±1.03)‰ when the concentrations of propofol were 7.5mg/kg,25.0mg/kg,and 50.0mg/kg respectively.The rates of micronuleus in foetal liver were (2.39±1.26)‰,(3.01±1.88)‰,and(2.98±1.87)‰ respectively with the same concentrations as above.Both groups were negative.Conclusion Propofol has no any significant impairment on the chromosome in rats.
