摘 要:提出了应用静电离子色谱法 ( EIC)同时分离测定抗坏血酸、脱氢抗坏血酸和变异脱氢抗坏血酸的方法。在自制的 ( EIC)固定相表面上 ,由于存在着微螺旋状胶束和均匀分布的正、负电荷 ,使几种形态的抗坏血酸得到了很好的分离。使用示差 -折光检测器 ,测定抗坏血酸的线性范围为 0 .2~ 2 mg· m L- 1。维生素 C片溶液 ( 1 mg·m L- 1)在常温下被空气氧化而失活的速度为 0 .0 4 5 mg·m L- 1· d- 1。该方法克服了样品测试过程中 ,由于抗坏血酸的变异而引起的系统误差 ,可为生物活性物质的动态分析 。A new method for separating and determining ascorbic acid (AA), dehydroascorbic acid (DHAA) and dehydroisoascorbic acid (DHIAA) simultaneously using electrostatic ion chromatographys (EIC) was presented. Ascorbic acid and dehydroascorbic acid were separated completely due to the presence of the micro helicoid micelle and the static electrification of the positive charge and negative charge distributed uniformly on the surface of stationary phase made in our lab. The detector used in this work was differential index refractometer, and the linear range was shown to be 0.2~2 g·L -1 . The Linear regression equation was Y=0.054 X+0.003. The linear correlation coefficient was 0.999. The disintegration rate of ascorbic acid in the sample was found to be 0.045 g·L -1 ·d -1 . The systematic error could be eliminated in biological sample analysis. The method introduced in this paper would provide a novel tool for dynamic analysis of samples with biological activity and for studying reaction mechanism of organic acids.
