摘  要:目的 :研究重组人肿瘤坏死因子 (rhTNF)及其联合化疗药物的体外杀伤肿瘤细胞的作用。方法 :采用体外细胞毒方法 (MTT)及流式细胞仪分析法检测重组人肿瘤坏死因子或 /和阿霉素 (ADM)对大鼠Walker 2 5 6肿瘤细胞作用后的细胞毒性及细胞分裂周期各时象的变化。结果 :rhTNF有很强的抗瘤活性 ,与阴性对照组比较有显著差异 (P <0 .0 1) ,并有很好的量效关系 (Υ =0 .9811) ,与ADM联用表现有协同增强细胞毒性 ;rhTNF使G2、S期细胞减少 ,增殖指数 (PI)降低 ,与ADM联用 ,其作用更加显著。结论 :rhTNF与ADM联用对Walker 2 5 6肿瘤细胞有协同增强的杀伤作用。Objective: To investigate the effects of rhTNF alone and in combination with antitumor chemicals on Walker-256 cell line in vitro. Methods: the cytotoxic activity in vitro were be examined by MTT method and the flow cytometry(FCM) analysis and alteration of every phase of cell cycle for rhTNF or (and) Adriamycin(ADM) against Walker-256 cell line. Results: rhTNF had a strong activity of antitumor in vitro and good dose-effect relationship(r=0.9811). Compared with negative control group, the effect of rhTNF was significant (P<0.01); rhTNF with ADM had a synergistic enhancement of antitumor activity in vitro. RhTNF alone reduced s-phase and G2-phase cells, meanwhile, Proliferation Idex (PI) declined, combining with ADM, this kind of effect was more significant. Conclusion: rhTNF in combination with ADM had a synergististic enhancement of antitumor activity in vitro.
