摘  要:目的 :研究东亚钳蝎毒抗癌多肽 (APBMV)对人早幼粒白血病细胞系HL 6 0和人肝癌细胞系SMMC 772 1及对小鼠肝癌H2 2 移植瘤的作用。方法 :体外应用MTT法、生长抑制实验和集落形成实验 ,观察APBMV对HL 6 0、SMMC 772 1及H2 2 瘤细胞的作用 ;体内实验采用小鼠H2 2 实体瘤模型 ,检测肿瘤生长抑制率 (IR)、白细胞计数(WBC)和脾脏指数 (SI) ,观察APBMV对H2 2 带瘤小鼠的影响。结果 :APBMV对HL 6 0和SMMC 772 1细胞均具较强的毒性作用 ,其IC50 值分别为 10 .74 μg ml和 11.33μg ml;APBMV可显著抑制两种细胞的生长 ,剂量效应关系明显 ,HL 6 0和SMMC 772 1细胞 2 4h、4 8h和 96h的IC50 值分别为 19.4 1μg ml、9.90 μg ml、11.4 1μg ml和l5 .87μg ml、13.0 5μg ml、8.70 μg ml;APBMV浓度 >8μg ml时 ,可明显抑制SMMC 772 1细胞的集落形成 (P <0 .0 1) ,IC50 为 10 .83μg ml。H2 2 细胞经APBMV处理后 ,代谢MTT的能力明显低于对照组 ,但剂量效应关系不明显 (P >0 .0 5 )。APBMV能显著抑制H2 2 带瘤小鼠的肿瘤生长 ,IR最高达 4 0 .30 % (P <0 .0 1) ,小鼠WBC和SI无明显变化或略高于对照组 ,而 5 Fu组小鼠WBC和SI均明显下降 (P <0 .0 1)。结论 :APBMV是东亚钳蝎蝎毒中有效低毒的抗肿瘤成分 ,对HL 6 0、SMMCPurpose:To investigate the effect of antineoplastic polypeptide from Buthus martensii venom (APBMV) on the cultured human promyelocytic leukemia cells HL 60 and hepatoma cell line SMMC 7721 and hepatoma H 22 bearing mice.Methods:MTT colorimetric method, growth inhibiting test and colony formation assay were used in the in vitro test. H 22 bearing mice were applied in the in vivo experiment. Through measuring tumor growth inhibitory rate(IR),white blood cell (WBC) number and spleen index (SI) ,we explored the influence of APBMV on H 22 bearing mice.Results:APBMV possessed stronger cytotoxicity on HL 60 cells and SMMC 7721 cells, and IC 50 was 10.74 μg/ml and 11.33 μg/ml , respectively. APBMV could dramatically inhibit their growths. There were obvious dosage response correlations. The IC 50 of HL 60 and SMMC 7721 at 24h, 48h and 96h were 19.41 μg/ml, 9.90 μg/ml, 11.41 μg/ml and 15.87 μg/ml, 13.05 μg/ml, 8.70 μg/ml, respectively. When the concentration of APBMV exceeded 8 μg/ml, the colony formation rate of SMMC 7721 cells decreased dramatically ( P <0.01).Dosage effect relationship was evident, and IC 50 was 10.83 μg/ml . The ability to metabolize MTT of H 22 cells was lower than control distinctively after the cells were treated by APBMV, whereas dose response relationship was not clear(P>0.05).Tumor growth of H 22 bearing mice was markedly inhibited by APBMV,the growth inhibiting rate was reached 40.30% ( P <0.01). After treatment by APBMV , white blood cell(WBC) number and spleen index of H 22 bearing mice were not changed markedly or were slightly higher than control group. In 5 fluorouracil treated group, WBC number and spleen index of H 22 bearing mice were all reduced markedly( P <0.01). Conclusions:APBMV was one kind of effective and low toxicity antineoplastic components in the venom of Buthus Martensii Scorpion.
