摘 要:目的 :研究IL 18对人卵巢癌细胞系HO 8910的抗肿瘤免疫作用。方法 :ELISA法测定IL 18诱导的外周血单个核细胞 (PBMC)培养上清液中IFN γ浓度 ;MTT法测定PBMC的杀伤活性 ;流式细胞仪测定HO 8910的细胞周期分布。结果 :不同浓度IL 18(0、2、4、6、8、10 μg ml)能较好诱导PBMC分泌不同水平的IFN γ(0、0 .39、0 .6 6、0 .81、1.5 6、1.5 8IU ml) ;PBMC与不同效靶比的肿瘤细胞共同温育后 ,IL 18组PBMC对肿瘤细胞杀伤率较对照组为高 (P=0 .0 16 ) ;流式细胞仪检测表明IL 18使HO 8910卵巢癌细胞的S期百分比明显下降 ,分别由 31.2 6 %降为 5 .18%(上清液 ) ,2 9.0 1%降为 18.80 % (上清液和PBMC)。结论 :IL 18能诱导PBMC分泌IFN γ因子 ,IL 18能提高PBMC对HOPurpose:Antitumor effect of IL 18 on ovarian carcinoma cell line was assessed.Methods:Measurement of IFN γ concentration in the supernatant of IL 18 induced PBMC by ELISA method. Tumoricidal activity of PBMC was measured by MTT method. The cell cycle distribution of HO 8910 was measured by flow cytometry.Results:With the increase of IL 18 concentrations (0,2,4,6,8,10 μg/ml), the IFN γ level at supernatant were correlatively increased(0,0.39,0.66, 0.81,1.56, 1.58IU/ml).After co culture with tumor cells at different ratio, tumoricidal activity of PBMC was higher in IL 18 group ( P =0.016).S phase percentage of HO 8910 ovarian carcinoma was decreased from 31.26% to 5.18% in the supernatant without PBMC and from 29.01% to 18.80% in the supernatant with PBMC. Conclusions:IL 18 could induce the secretion of IFN γ. It could not only enhance the tumoricidal activity of PBMC to HO 8910 ovarian carcinoma cell line, but also inhibit the growth of ovarian carcinoma.
