穿心莲内酯体外稳定性研究

摘　　要：目的 :考察穿心莲内酯在体外的稳定性 ,为制剂研究提供有效的参考。方法 :将穿心莲内酯原料药分别溶解于不同pH值的磷酸盐缓冲液中 ,采用经典恒温法 ,用反相高效液相色谱法测定穿心莲内酯的含量 ,并在此基础上考察该药 37°C时在不同生物样品中的稳定性和该药在不同有机溶剂中的稳定性。结果 :穿心莲内酯在不同 pH值条件下稳定性不同 ,以pH3～ 5最为稳定 ;而生物样品中 ,在小牛血清中最稳定 ,小鼠肺匀浆中次之。结论 :穿心莲内酯在不同条件下其稳定性不同 ,该结果为穿心莲内酯及其相关制剂的研究提供一定的参考。Objective: In order to provide an effective reference to the study on its preparation, the stability in vitro of andrographolide was investigated. Methods: Andrographolide was dissolved in phosphate buffer solutions of different pH values; classical isothermal method was employed; the content of andrographolide was determined by RP HPLC; then the stability of the drug in different biological media and organic solvents at 37°C was investigated. Results: The stability of andrographolide was different at different pH values and was best in the condition of pH=3～5. In biological media, it was most stable in bovine serum and the next was in mice homogenate. Conclusion: In different conditions, the stability of andrographolide was different. It provided a reference to the study on andrographolide and its preparation.
