摘  要:目的 :观察p2 7蛋白表达水平与肾癌GRC - 1细胞端粒酶活性的关系及对其诱导凋亡的作用。方法 :采用脂质体介导的方法将 p2 7质粒导入GRC - 1细胞中 ,应用TRAP -PCR -ELISA、电镜及Tunel等技术检测外源性 p2 7蛋白对GRC - 1细胞端粒酶活性、细胞凋亡的影响。结果 :转染 p2 7质粒的GRC - 1细胞端粒酶活性显著下降 ,伴有细胞凋亡、细胞生长停滞。结论 :外源性p2 7基因的表达可使GRC - 1细胞端粒酶活性降低 ,细胞生长停滞 ,诱发细胞凋亡。提示细胞端粒酶活性的改变与细胞周期的调控密切相关。Objective:To observe the effect of exogenous p27expression on GRC-1 cells and to investigate the relationship between p27 expression and cellular telomerase activity.Methods:p27 cDNA was introduced into GRC-1 cells by using lipofectin mediation; By means of TRAP, electromicroscope and Tunel,the changes of GRC-1 cells in telomerase activity and cell apoptosis were detected.Results:A significant association was observed between increased expression of p27 and decreased telomerase activity. Compared with control,GRC-1/pCMV-p27 presented a significant drop in telomerase activity expression,accompanied by remarkable apoptosis and growth arrest.Conclusions:Exogenous p27 protein over-expression promotes apoptosis of GRC-1 cells and reduces telomerase activity and can inhibit cell proliferation.These indicate that controlling the cell cycle correlated with the changes of telomerase activity.
