摘  要:目的 :探讨三氧化二砷 (As2 O3 )对人卵巢癌细胞株 3AO细胞裸鼠皮下移植瘤生长的抑制作用及作用机制。方法 :4× 10 6个 3AO细胞接种于裸鼠皮下 ,采用不同浓度As2 O3 腹腔注射治疗 ,与顺铂 (cDDP)进行对比 ,计算移植瘤的体积、瘤重、抑瘤率及Fas、FasL基因表达水平的变化。结果 :As2 O3 对移植瘤生长的抑制作用明显优于cDDP ,且具有剂量低赖性 ;As2 O3 作用后Fas基因的表达呈升调节 ,而FasL基因的表达无明显变化。结论 :As2 O3 对人卵巢癌移植瘤的生长具有明显的抑制作用 ,其机制与Fas基因的过度表达有关。Objective:To explore the inhibitory effect of arsenic trioxide (As_2O_3) on the growth of 3AO cell subcutaneous implanted tumor and its mechanisms. Methods: Nude mice were subcutaneously implanted with 4×10 6 3AO cells,then treated with intraperitoneal injection of different concentration of As_2O_3 and cDDP, and the volumes of implanted tumors, the tumor inhibition rates, Fas gene and FasL gene expressions were calculated. Results:Compared with cDDP, 3AO cell implanted tumor inhibition rates by As_2O_3 were significantly higher in dose-dependent manner; As_2O_3 up-regulated Fas gene expression, but did not affect FasL gene expression. Conclusions:As_2O_3 can inhibit the growth of human ovarian carcinoma implanted tumor, and its mechanisms are related to Fas gene over-expression.
