摘 要:采用免疫组织化学方法及放射配体结合分析法 ,观察清开灵及侧脑室注射c fos反义寡核苷酸阻断c fos基因表达后脑组织蛋白激酶C(PKC)表达量和Glu的N_甲基_D_天门冬氨酸 (NM DA)受体的数目、亲和力变化的影响。结果 :清开灵可有效地抑制Glu所致的脑组织c fos蛋白及PKC蛋白的表达增强 ,并下调Glu的NMDA受体数目 ;而c fos反义及正义寡核苷酸对Glu的NMDA受体数目及Glu所致的脑组织PKC蛋白表达增多均无明显影响。结论 :清开灵抗Glu神经毒性的脑保护作用的机制与其抑制cThe effects of Qingkailing Injection (QI) on c fos gene expression in relieving neurotoxic brain injury induced by glutamate were studied in rats by the immunohistochemical method and radioligand binding assay. The effects of QI and of the blockage of c fos gene expression by the right ventricle injection of c fos antisense oligonucleotide on the amount of cerebral protein kinase C (PKC) expression, and on the amount and affinity of N methyl D aspartic acid (NMDA) receptor of glutamate were also studied by the methods as same as the above mentioned. The results showed that QI could effectively inhibit the increase of the expression of cerebral c fos protein and PKC protein induced by glutamate, and lower the amount of NMDA receptor of glutamate; while both c fos antisense oligonucleotide and c fos sense oligonucleotide had no marked effects on the amount of NMDA receptor of glutamate and the increase of the expression of cerebral PKC protein induced by glutamate. The results suggest that the mechanism of the protective effects of QI against glutamate induced cerebral injury is related to its inhibiting the expression of c fos protein and PKC protein and lowering the amount of NMDA receptor.
