摘 要:目的 通过观察激素对胸腺细胞凋亡及Fas表达的影响 ,探讨糖皮质激素、雌激素、甲状腺素在重症肌无力(MG)发生、发展和治疗中的作用。方法 用流式细胞术分析胸腺细胞Fas、CD4 和CD8分子的表达 ;用琼脂糖凝胶电泳和细胞形态学观察细胞凋亡 ;用噻唑蓝 (MTT)比色分析法观察细胞的增殖情况。结果 地塞米松和雌激素均能抑制MG患者胸腺细胞增殖 ,对胸腺细胞的细胞增殖抑制率分别为 63.72 %和 61 .0 6% ,胸腺细胞DNA电泳可见梯状条带。甲状腺素可促进胸腺细胞增殖。地塞米松诱导MG患者胸腺细胞Fas表达百分率增加 ,数据为 (2 0 .38±6 .0 7) % ,与空白对照组 (1 2 .43± 4.32 ) %相比 ,差异有显著性(P<0 .0 5)。Objective To study the role of glucocorticoid,estrogen and thyroxine in Fas molecular expression and apoptosis of thymocytes in patients with myasthenia gravis.Methods Expression of the thymocytes Fas,CD 4 and CD 8 molecules was detected by using flow cytometry.The thymocytes apoptosis was observed by agarose gel electrophoresis and cell morphology.The cells proliferation was studied by MTT analysis methode.Results Both estrogen and dexasone could inhibit the proliferation of thymocytes of MG patients with the difference being significant (P<0.01) between experiment groups and control groups.The cultivated cells with dexasone showed character of apoptosis and the DNA of them showed typical ladder appearance in electrophoresis.The proportion of Fas thymocytes only in dexasone groups was significantly higher (P<0.05) than controls.Thyroxine could accelerate the proliferation of thymocytes of MG and normal human with the difference being significant (P<0.05) between experiment groups and control groups.Conclusion The hormones could obviously affect the proliferation and apoptosis of thymocytes in patients with myasthenia gravis.
