摘 要:目的 :建立一种准确、可靠的白血病免疫分型检测方法。方法 :采用流式细胞仪单染色和三色荧光标记技术。结果 :单染色标记法检测 4 8例白血病患者 ,诊断不明确 9例 (18.7% ) ,初始诊断与最后诊断不符 2例 (4% ) ,三染色标记法检测 36例白血病患者 ,未发现诊断不明确者或初始诊断与最后诊断不符者 ;且通过二维表型图谱 ,可发现相关抗原共同表达 ,利于混合型白血病诊断。结论 :流式细胞仪三色荧光标记法进行白血病免疫分型 ,克服了单染色标记法细胞分群不清的问题 ,对白血病诊断、发生发展和预后判断有重要的指导意义。Objective:To establish a accurate and reliable method in examining immunophenotypes of leukemia.Methods:To use mono color and three color fluorescent staining methods by flow cytometry.Results:Of the 48 cases detected by mono color method,nine cases (18.7 %) couldn't be diagnosed definitely and two cases (4 %) were inconsistent with the final diagnosis.For the other 26 cases detected by using three color method,however,none couldn't be diagnosed definitely or were inconsistent with the ultimate diagnosis.Meanwhile,co expression of related antigens detected by two dimension maps was diagnosed mixed leukemia.Conclusion:As a accurate and reliable method,three color fluorescent staining method can overcome the difficulties in cyto grouping encountered by mono color method and it is extremely important to diagnosis,development and prognosis of leukemia.
