半相合骨髓移植的动物实验研究

摘　　要：目的 :观察半相合骨髓移植对 GVHD或 GVL的影响。方法 :以 C57BL - 6小鼠急性放射病为模型 ,进行半相合小鼠骨髓移植 ,同时设异基因骨髓移植为对照 ,观察移植后小鼠存活率、造血重建、GVHD或 GVL作用。结果 :GVHD中 ,半相合骨髓移植组 30天死亡率低于基因骨髓移植。半相合骨髓移植与异基因骨髓移植比较 ,外周血象或骨髓 CFU - GM恢复快 ,以 9天以后明显 ,脾结节数及嵌合体测定高于异基因组。体内混合淋巴细胞反应证实半相合骨髓移植较异基因骨髓移植 GVHD小。在 GVL中 ,异基因骨髓移植组主要死于 GVHD。半相合骨髓移植组 30天生存率仍为 5 6 .3% ,表现出明显 GVL作用。结论 :半相合骨髓移植在减轻 GVHD同时 ,产生明显的 GVL作用。Objective:To observe the effects of half matched BMT on GVHD and GVL.Methods:Half matched BMT was performed in C 57 BL 6 murine model of acute radiation disease to observe the survival rate,hematopoietic recovery,GVHD and GVL in the mice,controlled with unmatched allogeneic BMT group.Results:For GVHD the mortality on day 30 in half matched group was lower than in the unmatched group.The hematopoietic recovery in half matched group was faster than in allo BMT group,esp.after day 9,as evidenced by the blood routine,bone marrow CFU GM assay,spleen node and chimera assay.In vivo mixed lymphocyte reaction test showed a weaker GVHD reaction in half matched group,compared to the allogeneic group.For the induction of GVL effects,the mice in allogeneic group were mainly died of GVHD while the half matched group showed considerable GVL effects with a survival rate of 56.3 %(9/16) on day 30.Conclusion:While lessening severity of GVHD,half matched BMT had considerable GVL effects.
