摘  要:目的 探讨视黄酸受体基因在人胚肾上腺和神经母细胞瘤中表达的意义。方法 应用原位分子杂交的方法 ,检测人胚肾上腺发育过程中视黄酸受体基因表达的水平 ,以及不同分化程度的神经母细胞瘤视黄酸受体基因的表达 ,并对表达程度进行半定量分析。结果 在胎儿肾上腺神经母细胞中 ,RARαmRNA、RARγmR NA明显表达并逐渐增强 ,1 9周龄后则急剧下降。RARβmRNA、RXRαmRNA表达稍弱 ,在 1 9周龄前基本保持同一水平。RXRβmRNA在 1 9周龄前一直是微弱表达。RXRγmRNA表达呈明显下降趋势。在胎儿肾上腺嗜铬细胞以及胎儿区和最终区的细胞中 ,6个mRNA均为微弱表达或基本不表达。在神经母细胞瘤细胞中 ,RAR类mRNA以及RXRαmRNA、RXRβmRNA在分化不良组中均明显表达 ,在分化良好组中表达减弱。两组相比均有显著性差异 (P <0 .0 1 )。RXRγmRNA在神经母细胞瘤中不表达。结论  (1 )RARα、RARγ可能参与了神经母细胞分化的介导 ;(2 )视黄酸受体基因表达水平与神经母细胞瘤的分化程度呈负相关 ;(3)孕 2 0周之前的肾上腺发育延滞可能与神经母细胞瘤的发生有关。Purpose: To explore the relationship between retinoic acid receptors and development of human embryonal adrenal and tumorigenesis of neuroblastoma. Methods: The expressions of retinoic acid receptors messenger RNA in human embryonal adrenal during development and in human neuroblastoma with different pathologic classification (favorable and unfavorable) were detected by in situ hybridization, and the levels of the expression were semiquantitated. Results: RARα mRNA and RARγ mRNA were both expressed abundantly in fetal adrenal neuroblastic cells. The intensity of expression of these two receptors was significantly increased with the process of gestational weeks, but was abruptly reduced after 19 weeks' gestation. Expressions of RARβ mRNA and RXRα mRNA in neuroblastic cells were low during 13 - 19 weeks' gestation, and expression of RXRβ mRNA was weak during those times. Expression of RXRγ mRNA was high in 13 week's gestation, then was significantly reduced with the development of neuroblastic cells. Expressions of RARs mRNA and RXRs mRNA were weak or almost undetectable in chromaffin cells, fetal zone, and definitive zone throughout the development. In neuroblastoma cells the expression levels of RARs mRNA, RXRα mRNA and RXRβ mRNA were all significantly higher in unfavorable group than in favorable group (P<0.01). Expression of RXRγ mRNA was not existent in neuroblastoma cells. Conclusions: This study suggests that: (1) RARα and RARγ are likely to play major roles in mediating the differentiation of neuroblastic cells; (2) There is a reverse change between the expression levels of retinoic acid receptors and the differentiation degrees of neuroblastoma cells; (3) Retarded differentiation of embryonal adrenal neuroblastic cells before 20 weeks' gestation may be related to the tumorigenesis of human neuroblastoma.
