核因子-κΒ在眼内炎症反应及视网膜增生性病变中作用的初步实验研究

摘　　要：目的 :探讨核因子 (nuclearfactor,NF) κB活性对眼内炎症反应以及视网膜增生性病变发病的影响。方法 :10只健康新西兰大白兔 ,右眼造成视网膜裂孔后随机等分两组 ,对照组右眼玻璃体内注入白细胞介素 (interleukin ,IL) 1β 10 0ng/10 0 μl,治疗组右眼于注入等量IL 1β前 1小时先注入NF κB的特异性抑制剂二硫代氨基甲酸乙酯 (pyrrolidinedithiocarba mate,PDTC) 10nmol/ 10 μl。裂隙灯和间接眼底镜观察兔眼 1个月。 1月后取右眼行光镜检查。结果 :观察期间对照组 2只兔眼玻璃体中度混浊、3只重度混浊 ,治疗组 3只兔眼玻璃体轻度混浊、2只中度混浊。 1个月时 ,对照组 4只眼视网膜前有增生物形成 ,治疗组无类似改变。组织学检查证实对照组视网膜前增生物形成。结论 :NF κB参与视网膜损伤和IL 1β诱导的兔眼眼内炎症反应及视网膜增生性病变的发生 ;抑制NF κB活性可能对葡萄膜 -视网膜炎。Objective:To explore the effect of the activity of nuclear factor (NF) κB on the development of intraocular inflammation and proliferative retinopathy.Methods:Ten healthy New Zealand rabbits were randomly divided into two groups after a retinal break was made in the right (experimental) eye.Interleukin (IL) 1β 100 ng/100 μl was injected intravitreally in the control group,and pyrrolidine dithiocarbamate (PDTC) 10 nmol/10 μl was injected intravitreally one hour before IL 1β injection in the treatment group.Experimental eyes were followed one month with slit lamp and indirect ophthalmoscope,and at the end of one month,the eyes were enucleated and studied with light microscope.Results:Of the control group,moderate vitreous opacity was found in 2 eyes,and severe in 3 eyes,while of the treatment group,vitreous opacity was mild in 3 eyes and moderate in 2 eyes.Pre retinal proliferation was present in 4 eyes in the control group,but none in the treatment group,which was confirmed by histopathological study.Conclusions:NF κB is involved in mediating intraocular inflammatory reaction and proliferative retinopathy induced by retinal damage and intraocular injection of IL 1β in rabbits′ eyes.Inhibition of NF κB activation may have therapeutic value for inflammation associated retinal diseases such as uveo retinitis and proliferative retinopathy.
