This study tested the hypothesis that heat stress respond is associated with the change in rat skeletal muscle with single-bout exercise. Male rats were assigned to one of the following four experimental groups (n = 10/group): (1) normothermic sedentary (Norm Sed), (2) heat stress (Heat Sed), (3) normothermic exercise (Norm Exe), or (4) heat stress before exercise (Heat Exe). Animals in the Heat Sed and Heat Exe groups were exposed to 15 min of whole body hyperthermia (colonic temperature 41℃~41.5℃). Twenty-four hours after heat stress respond, both the Norm Exe and Heat Exe groups were subjected to single-bout exercise. After exercise, the animals were anesthetized, and the gastrocnemius were removed, and SOD, MDA and the relative levels of heat shock protein 72 (HSP72)were analyzed. Compared with Norm Sed, the relative content of HSP72 in the gastrocnemius in Heat Sed was significantly elevated (P < 0.05). After heat stress, the content of MDA in the gastrocnemius in Heat Exe significantly decreased (P < 0.05) and the content of SOD increased (P > 0.05) ,as compared with that in Norm Exe. These data demonstrate that heat stress respond before single-bout exercise can reduce the content of MDA in gastrocnemius.
