摘  要:目的··:观察可卡因 -苯丙胺调节转录肽 (CART)mRNAs在慢性吗啡处理后大鼠的下丘脑中表达的变化。方法·· :采用PT -PCR克隆CART基因 ,用原位杂交观察慢性吗啡依赖大鼠下丘脑中CARTmRNAs的表达。结果··:在视上核 (SON)和室旁核 (PVN) ,正常组的CARTmRNAs阳性细胞数分别为44.2±s14.3和85.3±s34.6 ,慢性吗啡处理组分别为63.8±s18.2和118.5±s55.0 ;纳洛酮可阻断慢性吗啡处理引起的CARTmRNAs阳性细胞数的增加,与吗啡依赖组相比分别减少34.0 %和58.6 %。结论·· :CART可能参与吗啡依赖的形成;由于中枢CART的增加可明显抑制动物的进食行为 ,降低动物的体重 。Objective: To observe the changes of CART mRNA expression in hypothalamus in morphine-dependent rats. Methods: With RT-PCR and in situ hybridization (ISH), expression of cocaine-amphetamine-regulated transcript (CART) mRNA in hypothalamus of rats treated with chronic morphine were investigated. Results: After chronic administration of morphine, the numbers of CART mRNA positive neurons in the supraoptic nucleus (SON) and the paraventricular nucleus (PVN)(63.8±s 18.2 and 118.5±s 55.0)increased,compared with those (44.2±s 14.3 and 85.3±s 34.6) of the control group, but the withdrawal induced by naloxone blocked the alteration caused by chronic morphine treatment. Conclusion: The present results suggested that CART might be involved in the development of morphine dependence; the increases of expression of CART mRNA in hypothalamus might contribute to the reduce of feeding and loss of weight in the development of morphine dependence since elevation of CART in brain inhibited the behavior of feeding and resulted in the weight loss.
