粘液表皮样癌细胞中脆性组氨酸三聚体基因异常表达的研究

摘　　要：目的 :检测粘液表皮样癌细胞中FHIT基因的异常表达。方法 :应用逆转录巢式聚合酶链反应 (RT -PCR)及DNA测序技术检测粘液表皮样癌和粘液表皮样癌细胞中FHIT基因表达情况。结果 :粘液表皮样癌细胞系细胞中存在FHIT基因部分缺失 ,产生异常转录本 ,大小约 2 47bp .FHIT基因mRNA异常转录本为多个外显子缺失所致 ,E1-E8全部缺失。结论 :FHIT基因在粘液表皮样癌细胞中的异常表达可能在粘液表皮样癌的发生和发展中起作用。AIM:To determine whether the FHIT gene at 3p14.2 is altered in mucoepithemoid carcinoma cell line(MEC-1)cells. METHODS: Total RNA of MEC-1 cells was extracted. The deletions within the FHIT gene were detected by RT-PCR and DNA sequencing technologies. RESULTS: Abnormal sized FHIT gene transcript was detected in the MEC-1 cells, about 247bp. The sequence analysis of the aberrant cDNAs revealed deletions of exon 1-8 and exon 10. CONCLUSION: The deletion of the FHIT gene in mucoepidermoid carcinoma might support the hypothesis that the FHIT gene alteration is involved in tumorigenic development of mucoepidermoid carcinoma.
