摘　　要：目的　观察 IL - 2对 C6大鼠星形神经胶质瘤细胞增殖的影响 ,并从蛋白及 m RNA水平观察白细胞介素 - 2受体(IL - 2 R)α,β,γ 3条链在 C6神经胶质瘤细胞中的表达 .方法　应用细胞培养、MTT法、3H- Td R掺入法、免疫细胞化学方法和 RT- PCR技术 .结果　 1IL - 2 (1× 10 4 ～ 1× 10 6 U·L- 1 )可显著促进 C6细胞的增殖 ;2 C6细胞中检测到 IL - 2 R的 β、γ2条链的免疫反应阳性物质 ;3IL- 2 Rβ,γ m RNA在C6细胞中均有表达 .结论　 IL - 2可显著促进 C6神经胶质瘤细胞的增殖 ,C6细胞表达 IL- 2 Rβ、γ2条链的蛋白与 m RNA,可能介导 IL- 2增殖信号的传递 ,从而初步证实 IL- 2可直接作用于神经胶质细胞并进一步阐明了 IL- 2增殖信号的转导机制 ,为揭示细胞因子在神经系统中的普遍作用机制提供更多证据 .AIM To investigate the effects of interleukin 2 (IL 2) on the proliferation of C6 glioma cell line and the expressions of α, β, γ chains of interleukin 2 receptor (IL 2R) in the cell line at protein and mRNA levels. METHODS Techniques used in this experiment included cell culture, MTT, 3H TdR incorporation,immunocytochemistry and RT PCR. RESULTS ① IL 2 significantly increased the proliferation rates of C6 cells; ② IL 2Rβ, γ immunoreactive production was detected in C6 cells; ③ Il 2Rβ and IL 2R γ mRNA were both expressed in the cells. CONCLUSION IL 2 could stimulate the proliferation of C6 cell line, also the protein and mRNA of IL 2Rβ, γ were expressed in the cells . All of them can provide more evidence for the direct action of IL 2 in glioma cells and further disclose the cytokin's role in the central nervous system.
