胃癌组织FasL表达上调与肿瘤浸润性淋巴细胞凋亡的关系

摘　　要：目的　探讨胃癌FasL表达上调与肿瘤浸润性淋巴细胞 (TIL)凋亡的关系。方法　采用免疫组织化学S -P法 ,检测5 0例胃癌组织中FasL表达及TIL的数量 ,采用脱氧核糖核酸末端转移酶介导的缺口末端标记技术 (TUNEL) ,对 40例胃癌组织中凋亡的TIL及肿瘤细胞进行观察。结果　 5 0例胃癌组织FasL蛋白表达程度不等 ,FasL表达程度高的组织的TIL计数低于FasL表达低的组织 (P <0 .0 5 ) ,同时其TIL凋亡指数高于FasL表达低的组织 ,而胃癌细胞的凋亡指数低于FasL表达程度低的组织 (P <0 .0 1) ,TIL凋亡指数与胃癌细胞的凋亡指数呈负相关 (γ =-0 .5 2 ,P <0 .0 1)。结论　人类胃腺癌有不同程度的FasL表达 ,其表达程度与TIL的浸润程度和凋亡相关 ,可能是胃癌免疫逃避的重要机制之一。Objective To study whether the up regulation of FasL expression is associated with increased apoptosis of Tumor infiltrating Lymphocytes(TIL) in huaman gastric adenocarcinomas(GAC).Methods The expression of FasL protein and the number of TIL in 50 specimens of GAC were detected by immunohistochemitry.Using terminal deoxynuceotidyl transferase mediated dUTP nick end labeling(TUNEL),apoptitic cells were detected in situ in 40 GAC specimens.Results The expression of FasL protein was detected in all 50 GAC examined.In those FasL extensive expression,the numbers of TIL were lower than those FasL weak expression( P <0.05),and their apoptotic index(AI) of TIL was higher and the AI of tumor cell was lower than those FasL weak expression,respectively( P <0.01).The AI of TIL was reversely correlated with the AI of tumor cell(γ=-0.52, P <0.01).Conclusion These data suggest GAC express FasL and its expression level associate with TIL infiltrating and apoptosis,this maybe a mechanism of immune evasion in GAC.
