MMP-2和TIMP-2在宫颈腺癌中的表达及与其转移的关系

摘　　要：目的　研究宫颈腺癌组织中基质金属蛋白酶 (MMP)和金属蛋白酶组织抑制因子 (TIMP)的表达及其与肿瘤浸润转移的关系 ,揭示宫颈腺癌的浸润转移机制。方法　应用免疫组化方法 ,检测了 10例慢性宫颈炎和 63例宫颈腺癌组织中MMP -2和TIMP -2的表达情况。结果　MMP -2和TIMP -2在慢性宫颈炎组织中均无阳性表达 ,而在宫颈腺癌组织中阳性表达率分别为 73 .0 % (4 6/63 )和 82 .5 % (5 2 /63 ) ,有显著性差异。MMP -2在腺型腺癌和乳头状腺癌组织中阳性表达率分别为 81.8% (18/2 2 )和 5 8.3 % (14 /2 4) ,无显著性差异 ,而MMP -2和TIMP -2表达与其临床分期也无相关性 (P >0 .0 5 )。MMP -2和TIMP -2表达间有非常显著相关性 (P <0 .0 1) ,同时MMP -2阳性表达与该肿瘤转移呈正相关 (P <0 .0 5 )。结论　MMP -2可作为预测肿瘤浸润和转移的 1个指标。Objective To investigate the expressions of matrix metalloproteinase 2(MMP-2) and tissue inhibitor of matrix metalloproteinase 2(TIMP-2) protein in the tissue of cervical adenocarcinoma and its correlation the metastasis of cancer and to reveal the mechanism of infiltration in this cancer.Methods The expressions of MMP-2 and TIMP-2 protein were observed by immunohistochemistry(IHC)technique in 10 cases of cervical chronic inflammation and 63 cervical adenocarcinoma.Results No positive expressions of MMP-2 and TIMP-2 in cervical chronic inflammation,while the positive rate was73.0% and 82.5% respectively in cervical adenocarcinoma.The positive rate of MMP-2 protein expression in cervical adenoferous carcinoma and papillary adenocarcinoma was 81.8% and 58.3% respectively.MMP-2 and TIMP-2 protein′s expressions had no relevance with clinical stage( P > 0.05 ).In this study there was a very significant correlation between the expressions of MMP-2 and TIMP-2 protein( P <0.01) and the expression of MMP-2 was positively correlated to metastasis of cancer( P <0.05).Conclusion The MMP-2 protein may act as an index to indicate the process of the infiltration and metastasis in cervical adenocarcinoma.
