甲基维生素B12对大鼠自身免疫性脑脊髓炎的治疗及溶血磷脂酸水平的影响

摘　　要：研究实验性自身免疫性脑脊髓炎 (EAE)及甲基维生素B12治疗中溶血磷脂酸 (LPA)水平的变化。通过注射豚鼠脊髓匀浆和完全弗氏佐剂诱导大鼠发生EAE。用有机溶剂提取、并进一步分离 ,最终用定磷方法测定甲基维生素B12治疗的EAE大鼠血浆及脑溶血磷脂水平 ,未经治疗的EAE大鼠为对照组。发现甲基维生素B12治疗组EAE大鼠与未经治疗EAE大鼠比较 ,恢复期脑与血浆LPA水平增高 ,分别为 30 2 3± 11 5 9μmol L血浆、2 45 4 5±144 89nmol g脑和 9 2 7± 3 2 5 μmol L血浆、5 6 33± 5 6 6nmol g脑 (P <0 0 5 ) ;急性期血浆的LPA增高 ,分别为 10 0 5± 1 70 μmol L血浆和 1 87± 0 5 9μmol L血浆 (P <0 0 5 )。溶血磷脂酸可能参与自身免疫性脑脊髓炎髓鞘的恢复过程。Lysophosphatidic acid (LPA) is a multi-functional phospholipid messenger which has growth factor-like effect and plays a role in damage recovery. This study was taroeted to LPA level and its changes after treatment with Methylcobal which enhance the recovery of damaged myelin in rat with experimental auto-immune encephalomyelitis(EAE). EAE was induced by injection of Complete Freund's adjuvant with spinal cord homogenate of guinea pig. Both cerebral tissue LPA and plasma LPA were assayed in EAE rats and Methylcobal injected EAE rats. The analysis was conducted by measuring the inorganic phosphorus component of LPA after extraction and isolation. The results showed that the level of plasma LPA of EAE rats injected with Methylcobal was higher than control (P<0.05) in acute phase(10.05±1.70μmol/L and 1.87±0.59 μmol/L respectively). The level of plasma and cerebral LPA of EAE rats injected with Methylcobal was also significantly higher than control EAE rats (P<0.05) in restorative phase(30.23±11.59μmol/L ?245.45±144.89 n mol/g brain tissue and 9.27±3.25μmol/L?56.33±5.66 n mol/g brain (P<0.05) tissue respectively). Complete restoration ratio was higher in the group injected with Methylcobal than in the control group. Histopathological microscopy also confirmed that their damaged myelin was obviously restored. These results suggested that LPA played a role in myelin repairing promoted by Methylcobal in EAE.
