树突状细胞激活的肿瘤浸润性淋巴细胞体外对自体肝癌细胞杀伤活性的研究

摘　　要：目的 :探讨树突状细胞 (DC)激活的肿瘤浸润性淋巴细胞 (TIL)体外对自体肝癌细胞杀伤活性。方法 :从肝癌患者外周血获取 DC,应用粒 /巨噬细胞集落刺激因子 (GM- CSF)、白细胞介素 - 4 (IL- 4 )和肿瘤抗原激活 DC,然后用 DC来激活 TIL,观察 TIL 在体外对自体肝癌细胞的杀伤活性。结果 :DC激活的 TIL 具有很高的对自体肝癌细胞杀伤活性 ,杀伤率为 (89.39±3.0 5 ) % ,明显高于未经 DC激活的 TIL、CD激活的 T淋巴细胞和未经 DC激活的 T淋巴细胞对自体肝癌细胞的杀伤率。结论 :肝癌患者外周血 DC能诱导 TILObjective: To study the killing activity of tumor infiltrating lymphocytes (TIL) stimulated by dendritic cells (DC) on autogenous hepatocarcinoma cells in vitro Methods: DC was isolated from hepatocarcinoma patients' peripheral blood DC were stimulated by granulocyte/macrophage colony stimulating factor (GM CSF), interleukin 4 (IL 4) and tumor antigen Then TIL were stimulated by DC, and their killing activity on autogenous hepatocarcinoma cells in vitro were observed Results: TIL stimulated by DC had very high killing activity on autogenous hepatocarcinoma cells and the killing rate was (89 39±3 05)% The killing rate was higher obviously than that of TIL not stimulated by DC and T lymphocytes stimulated by DC, or not on autogenous hepatocarcinoma cells, respectively Conclusions: The results indicate that DC from patients with hepatocarcinoma can induce efficient and specific anti hepatocarcinoma immune response
