阵发性睡眠性血红蛋白尿症患者T淋巴细胞对多向祖细胞的影响

摘　　要：将阵发性睡眠性血红蛋白尿症 (PNH)患者外周血制备的植物血凝素刺激的 T淋巴细胞条件培养液 (PHA-TCM)加入多向祖细胞的培养基中 ,用于 PNH骨髓混合集落形成单位 (CFU- Mix)的培养。发现在含有初发 PNH患者的 PHA- TCM培养条件下 ,骨髓的 CFU- Mix集落数明显减低。在含有治愈的 PNH患者的 PHA- TCM培养条件下 ,骨髓的 CFU- Mix集落数恢复正常。结果表明初发的 PNH患者In order to study the effect of T lymphocytes on pluripotential progenitors in the patients with paroxysmal nocturnal hemoglobinuria (PNH), phytohemagglutinin stimulating T lymphocytes conditioned medium (PHA LCM) prepared from PNH and normal blood was added to the culture medium of the hematopoietic progenitor culture for the culture of colony forming units granulocytes, erythrocyte, macrophage and megakaryocyte (CFU Mix) of the patients with PNH and normal. The results showed that the number of bone marrow CFU Mix in newly diagnosed PNH grown in the medium containing PHA TCM prepared from newly diagnosed PNH was obviously decreased as compared with that from normal, but the number of bone marrow CFU Mix in newly diagnosed PNH grown in the medium containing PHA TCM prepared from PNH in remission was significantly decreased as comapred with that from normal. The results suggested that the effect of T lymphocytes from PNH on pluripotential progenitors was declined.
