摘 要:目的 :研究核转录因子NF κB在人肝细胞肝癌组织中的表达及其与乙型肝炎病毒 (HBV)X蛋白的关系。方法 :用免疫组织化学S P法 ,检测 5 2例人肝细胞肝癌组织中核转录因子NF κB及HBVX蛋白的表达 ;用质脂体介导的基因转染法将HBVx基因真核表达载体pcDNA3 1 HBX转染入人肝癌细胞系HCC 92 0 4 ,检测肝癌细胞内核转录因子NF κB的表达。结果 :5 2例人肝细胞肝癌组织均有核转录因子NF κB的广泛表达 ,并且在 11例HBVX蛋白阳性的肝癌组织 ,核转录因子NF κB位于肝癌细胞胞质和胞核 ,而在 4 1例HBVX蛋白阴性的肝癌组织 ,核转录因子NF κB位于肝癌细胞的胞质。将HBVx基因真核表达载体 pcDNA3 1 HBX转染入人肝癌细胞系HCC 92 0 4 ,并在稳定表达X蛋白的肝癌细胞 ,核转录因子NF κB定位于其胞质和胞核 ,而未进行基因转染的亲体细胞 ,核转录因子NF κB仅定位于细胞质 ,细胞核无核转录因子NF κB的表达。结论 :核转录因子NF κB在人肝细胞肝癌组织中广泛表达 ,人肝细胞肝癌中存在着核转录因子NF κB的异常激活 ,并且核转录因子NF κB的异常激活与HBVX蛋白有关 ,X蛋白可激活核转录因子NF κB ,使其从细胞质转位于细胞核 ,这可能在HBV相关的人原发性肝细胞肝癌的发生中起一定作用。Purpose To investigate the relationship of the X protein of HBV and nuclear transcription factor κB (NF κB) and expression of NF κB in human hepatocellular carcinoma tissues. Methods Immunohistochemical S P method was used to detect expression of NF κB and the X protein of HBV in 52 cases of hepatocellular carcinoma. Gene transfection mediated by lipofectamine was used to transfect the eukaryotic expression vector pcDNA3 1 HBX of HBV x gene into human hepatocellular carcinoma cell line HCC 9204 and NF κB was detected. Results NF κB was widely expressed in human hepatocellular carcinoma tissues in total 52 cases and its expression was related to the X protein of HBV. NF κB was localized in both cytoplasm and nuclei of hepatocellular carcinoma cells in 11 cases with positive X protein of HBV while 41 cases were negative for X protein of HBV, NF κB was only localized in the cytoplasm of hepatocellular carcinoma cells. NF κB translocated to the nuclei of hepatocellular carcinoma cells after the eukaryotic expression vector pcDNA3 1 HBX was transfected into HCC 9204 cells, while in untransfected cells NF κB was localized only in cytoplasm. Conclusions NF κB is widely expressed in hepatocellular carcinoma tissues with different patterns based on the expression of the X protein of HBV. NF κB is abnormally activated in hepatocellular carcinoma, which is probably related to the X protein of HBV. The X protein of HBV can activate NF κB and make it translocate into the nuclei of hepatocellular carcinoma cells.
