摘  要:为探讨星形胶质细胞在癫痫发作中的作用 ,用肿瘤坏死因子 -α (tum or necrosis factor-α,TNF-α)刺激纯化培养的海马星形胶质细胞 ,将此条件培养基 (astrocytic conditioned m edium,ACM) 10 μl注入大鼠侧脑室 ,观察动物的行为、脑电图及杏仁核内谷氨酸 (glutamic acid,Glu)免疫组织化学反应。结果表明 ,侧脑室注射 ACM可引起大鼠癫痫样发作 ,脑电图出现阵发性痫样放电 ;杏仁核内 Glu免疫阳性反应增强 ,经多媒体彩色病理图文分析系统 (MPIAS)检测 ,阳性细胞面密度和平均光密度高于对照组。In order to explore the effect of astrocytes on epileptic seizure, TNF α(tumor necrosis factor α) was used to stimulate the purified cultural astrocytes and 10μl of the ACM was injected into lateral ventricle of rats. The animal behavior, EEG (electroencephalograph) and Glu (glutamic acid) immunohistochemistry reaction in amygdala were investigated . The result indicated that after injecting the ACM into the lateral ventricle, the seizure of the rat can be induced and the EEG showed epileptic discharges. In experimental group the Glu positive immunoreactivity was stronger than that in control one. Using MPIAS (multimedia pathology imaging analysis system) both the area density and optical density of positive cells were increased as compared with those in control one.This paper provided a direct evidence for the mechanism of epileptic seizure induced by astrocytes.\;
