大鼠肝癌发生过程中转化生长因子-β_1的表达及意义

摘　　要：为了进一步探讨转化生长因子 -β1 在肝癌发生中的作用和意义 ,本文采用免疫组织化学 ABC法 ,对二乙基亚硝胺(DEN)诱发大鼠肝癌发生过程中转化生长因子 -β1 (Transforming growth factor-β1 ,TGF-β1 )表达情况进行了观察。结果显示 :在正常大鼠肝脏 ,TGF-β1 的表达只局限于血窦内皮细胞或枯否细胞、汇管区血管内皮细胞及胆管上皮细胞 ,肝细胞呈TGF-β1 阴性表达。诱癌早期 (4～ 8周 ) ,大鼠肝小叶内除血窦内皮细胞或枯否细胞呈 TGF-β1 阳性表达外 ,可见少量散在分布的肝细胞呈 TGF-β1 阳性表达 ,阳性肝细胞胞质内可见棕褐色阳性反应颗粒。随着肝癌发展 ,TGF-β1 阳性肝细胞逐渐增多 ,至诱癌晚期 (18周 ) ,癌结节内的大多数肝癌细胞呈 TGF-β1 阳性表达。本研究显示 TGF-β1The expression of transforming growth factor β 1 (TGF β 1) during hepatocarcinogenesis induced by diethylnitrosamine (DEN) in rats was investigated with immunohistochemical ABC method.The results showed that the positive expression of TGF β 1 was detected in sinusoid endothelial cells or kupffer cells,the endothelial cells of blood vessels and bile ducts in portal areas,but not in hepatocytes in normal rat liver ; In the 4～8th week after DEN treatment, in addition to TGF β 1 positive sinusoidal lining cells ,a few TGF β 1 positive hepatocytes were detected in hepatic lobules .The TGF β 1 positive reactive products were brown, fine granules in the cytoplasm of these diffusely distributed hepatocytes. In the course of hepatocarcinogenesis,the number of hepatocytes expressing TGF β 1 increased progressively, and in 18th week after DEN treatment,the majority of the malignant hepatocytes in the neoplastic nodule expressed TGF β 1.The results suggest that the expression of TGF β 1 is related closely to the development and progression of hepatoma.
