摘　　要：目的 :探讨decorin对肝癌细胞株增殖及细胞凋亡的作用。方法 :用不同浓度的decorin和正常肝细胞株 (L 0 2细胞 )、肝癌细胞株 (SMMC 772 1、MM4 5 )共同孵育一定时间后 ,观察细胞的存活情况、形态学改变 ,用流式细胞仪分析DNA的分布。结果 :Decorin在体外能抑制肝癌细胞的增殖 ,诱导肝癌细胞出现典型的凋亡细胞特征 :细胞核浓缩、细胞膜皱摺、凋亡小体形成 ,而且其诱导凋亡作用呈剂量效应。结论 :Decorin作为一种生物制剂可诱导肝癌细胞凋亡。Objective To detect the effect of decorin on hepatoma cell lines in vitro.Methods Normal and hepatoma cell lines were cultured with a wide range concentration of decorin for various duration.Cell activity, morphologic changes and cell DNA distribution were studied.Results Apoptosis of hepatoma cell lines could be efficiently induced by decorin,with the presence of apoptotic changes in morphology,apoptotic peak before G 1 phase of cell cycles on FCM.Under the microscope,cells displayed a characteristic pattern of structural changes,including cell membrane chromatin condensation,nucleic fragmentation and apoptotic body formation.Apoptosis of these cells showed dose dependently.Conclusions Decorin can induce apoptosis of hepatoma cells.
