鲨鱼软骨制剂对人红白血病细胞增殖抑制作用的研究

摘　　要：目的 :研究鲨鱼软骨制剂 ( SCP)对人红白血病 ( K562 )细胞生长、抑制效应 ,探讨其抗癌作用机制。方法 :用不同浓度的 SCP加入体外培养的 K562细胞中 ,观察加药后细胞生长和有丝分裂指数 ( MI)的变化 ;用 MTT法检测其细胞毒作用 ;Hoechst3 3 3 4 2 /PI荧光双染色方法检测细胞凋亡。结果 :SCP明显抑制 K562细胞生长 ;IC5 0 值为 0 .7mg/m L ;MI于加药后 2 4h较对照组明显降低 ;凋亡细胞比例在用药后 48h达 44.55%。结论 :SCP可有效抑制人红白血病细胞的增殖。Objective:To study the effects and its mechanism of SCP on K562 cells proliferation.Methods:Using experiment in vitro, we added SCP of different concentrations into the medium in which the cells were cultrued.The changes of cell proliferation were assessed by MI and cell growth number. Cytoxicity rate was detected by MTT. The number of apoptotic cells was quantitatively assessed by fluorescent light microscope.Results:The cell growth was remarkably inhibited by SCP; the IC 50 values of SCP for K562 cells was 0.7 mg/mL.After 24 h of exposure to SCP, MI was remarkably reduced, the rate of apoptotic cells was increased.Conclusion:Malignant phenotype of K562 cells was changed and its proliferation ability was remarkably inhibited by SCP.
