摘 要:目的 探讨Fas和bcl- 2蛋白表达和DNA倍体异常在食管癌发生和发展中的作用。方法 应用流式细胞术和细胞免疫荧光技术对 70例食管鳞癌组织Fas、bcl- 2蛋白和DNA含量进行定量检测。结果 异倍体 36例 (5 1 4 % ) ,二倍体 34例 (48 6 % ) ;异倍体肿瘤其DNA指数、增殖指数和S期细胞比率均高于二倍体肿瘤 ,但差异无显著性 (P >0 0 5 ) ;Fas蛋白阳性表达 4 2例 (6 0 0 % ) ,bcl- 2蛋白阳性表达 6 5例 (92 9% ) ,Fas蛋白表达与肿瘤病理分级密切相关 (γ =0 5 84 ,P <0 0 5 ) ;异倍体肿瘤和二倍体肿瘤Fas和bcl- 2阳性表达率和表达量均差异无显著性 (P >0 0 5 )。结论 DNA倍体异常、Fas和bcl -Objective To explore the relationship among the Fas ,bcl-2 expression ,abnormal DNA ploidy and carcinogenesis of esophageal carcinoma.Methods Fas , bcl-2 protein expression and DNA content were quantitatively detected using flow cytomytry and cellular immunofluoresence technique in 70 cases of esophageal squamous cell carcinoma. Results 51 4% (36/70) of aneuploidy and 48 6% (34/70) of diploidy were observed in 70 cases of esophageal carcinoma. The DNA index, proliferation index and S-phase ratio in aneuploidy tumors were higher than those in diploidy tumors , but difference was not statistical significance(P>0 05). Positive expression rates of Fas and bcl-2 were 60 0% and 92 9% respectively and Fas increased along with the escalation of the pathological grading (γ=0 584,P<0 05). Fas and bcl-2 protein expression in aneuploidy tumors were lower than those in diploidy tumors , but difference was not significant(P>0 05). Conclusions DNA ploidy aberration,Fas and bcl-2 overexpression could play an important role in the development of esophageal carcinoma .
