摘  要:目的 :在细胞水平上精确定位解偶联蛋白 2 (uncouplingprotein 2 ,UCP2 )在肝脏组织中的表达 ,同时观察腹腔内注射细菌脂多糖 (lipopolysaccharide ,LPS)后不同时程UCP2 mRNA在肝脏中的表达 ,借此进一步探讨UCP2 在LPS引起的内毒素 (endotoxin ,ET)性发热中的作用。方法 :应用原位杂交 (in situhybridization ,ISH)技术和计算机图像分析技术检测能够反应小鼠肝脏中UCP2 mRNA表达的 3项重要参数值。结果 :LPS诱导肝细胞表达UCP2 mRNA ,使UCP2 mRNA在LPS致热小鼠肝脏中的表达显著高于同时期对照组的表达。结论 :在LPS引起的ET性发热中 ,LPS诱导肝细胞表达UCP2 mRNA ,使UCP2mRNA在肝脏中的表达增加。Objective: This study was designed to locate the expression of UCP 2 mRNA and to observe its expression in livers at different time points after intraperitoneal injection of lipopolysaccharide (LPS). Methods: Three important indices which can show the UCP 2 mRNA expression in livers of mice were measured by the method of in-situ hybridization and analyzed by using microscopic image quantitative analysis technique. Results: The LPS inducted the expression of UCP 2 mRNA in hepatocytes and improved the expression of UCP 2 mRNA in livers of feverish mice caused by LPS. The expression in feverish mice was much stronger than that of the control groups at the same time points. Conclusion: In endotoxic fever caused by LPS, LPS can induce the expression of UCP 2 mRNA in hepatocytes and improve its expression in livers of mice.
