端粒酶正义寡核苷酸对鼻咽癌细胞生长的作用

摘　　要：目的　探讨端粒酶正义寡核苷酸对鼻咽癌细胞生长的作用 .方法　脂质体介导端粒酶正义、反义及无义寡核苷酸转染鼻咽癌 CNE1和 CNE2 Z细胞 ,MTT法检测细胞增殖能力 ,流式细胞仪检测细胞周期 .结果　端粒酶正义寡核苷酸与反义寡核苷酸一样可抑制鼻咽癌 CNE1和 CNE2 Z细胞的生长 ,且呈剂量和时间依赖性 .1.5 μm ol· L- 1正义寡核苷酸与 CNE1细胞共培养 6 ,12 ,2 4和 4 8h时抑制率分别为16 .3% ,15 .6 % ,2 0 .2 %和 2 6 .3% ,浓度为 4 .5 μmol· L- 1时抑制率分别为 2 2 .5 % ,2 1.5 % ,32 .4 %和 39.9% ;在 CNE2 Z细胞 ,浓度为 1.5μmol· L- 1 时抑制率分别为 7.7% ,2 5 .2 % ,2 7.0 %和 2 8.8% ,浓度为 4 .5 μmol· L- 1时抑制率分别为18.1% ,30 .1% ,32 .8%和 32 .9% ;同时流式细胞仪检测到凋亡峰 ,含量分别为 2 5 .3%和 19.3% .无义寡核苷酸无此作用 .AIM To investigate the effects of telomerase sense oligodeoxynucleotide on the growth of nasopharyngeal carcinoma cells. METHODS Telomerase sense oligodeoxy nucleotide (SODN), antisense oligodeoxynucleotide (ASODN) and nonsense oligodeoxynucleotide (NODN) wereintroduced into nasopharyngeal carcinoma CNE1 and CNE2Z cells by using lipofectin mediation, the proliferation of cells was measured by MTT method, and cell cycle were measured with flow cytometer (FCM). RESULTS SODN inhibited the growth of CNE1 and CNE2Z cells as well as ASODN, and presented dose and time dependent manner. After being treated with 1.5 μmol·L -1 SODN for 6, 12, 24 and 48 h, the inhibition rates were 16.3%, 15.6%, 20.2% and 26.3% respectively in CNE1 cells, and 7.7%, 25.2%, 27.0% and 28.8% respectively in CNE2Z cells; with 4.5 μmol·L -1 SODN, the inhibition rates were 22.5%, 21.5%, 32.4% and 39.9% in CNE1 cells, and 18.1%, 30.0%, 32.8% and 32.9% in CNE2Z cells. Meanwhile, the apoptotic peaks were detected with FCM and the quantity were 25.3% and 19.3% respectively. NODN had no inhibitory effect on CNE1 and CNE2Z cells. CONCLUSION The telomerase SODN could inhibit the growth of nasopharyngeal carcinoma cells.
