摘 要:性别的DNA诊断具有一些优点并可补充或代替细胞遗传学诊断。作者试用未经纯化的微量绒毛DNA粗制液与生物素或地高辛配基标记的人Y特异DNA探针pHY2.1和cos84进行点杂交性别诊断。结果表明,未经抽提纯化的绒毛DNA可用于点杂交性别诊断;pHY2.1点杂交后男性与女性斑点显色强度有明显差异;cos84点杂交后男女斑点显色强度无明显差异;当绒毛DNA粗制品量为12ng时,与pHY2.1杂交后仍能得到明确的杂交信号。Sex determination by DNA technology has a series of merits and can replace or supplement cytogenetic diagnosis. It is especially important in the prevention of sex-linked genetic diseases such as Duchenne's muscular dystrophy (DMD), Lesch Nyhan's syndrome, and so on. We tried to diagnose the sex of fetus by using minute unpurified chorioic DNA, dot hybridized to biotinylated or digoxigenin-labeled human Y-specific DNA probes pHY2.1 and cos84. The results showed that unpurified chorionic DNA could be used for sex determination on dot hybridization; after dot hybridization with pHY2.1, distinct difference in signal degree between male and female DNA could be observed, while dot hybridization with cos84, there was no distinct difference of signal between the two sexes; clear hybridization signal could be obtained after hybridization with pHY2.1 when chorionic DNA was 12ng.
