摘  要:目的:利用基因表达谱芯片研究U937细胞在硫化砷作用前后基因表达的差异性。方法:采用急性单核细胞白血病细胞株U937,用 cy3和cy5两种不同荧光染料,通过逆转录反应将硫化砷处理前后的U937 mRNA分别标记成两种探针,并与载有一组靶基因的基因表达谱芯片进行杂交,经扫描、计算机软件分析寻找经硫化砷作用前后表达有差异的基因。结果:筛选出与细胞骨架、代谢相关酶和细胞信号转导等相关的表达有差异的基因共7条,其中1条表达上调,6条表达下调。结论:细胞骨架和细胞信号转导的改变可能参与硫化砷促进U937细胞凋亡的过程。Objective: To study the changes of gene expression of U937 cell treated with arsenic sulfide by the method of cDNA mi-croarray. Methods: mRNA of U937(menocytic leukimia cell lines) pre-or post-treated by arsenic sulfide was marked as two cDNA probe by two different dyes cy3 and cy5 . Meanwhile, they were hybridized with cDNA microarray and scanned for fluorescent intensity respectively. The genes of different expression were screened through the analysis of gene expression profile. Results: Seven genes were found out to be related to cell skeletal structure, enzymes of metabolism and signal transduction , which expressed differently after the treatment of Arsenic sulfide. One out of seven expressed as up-regulation and others as down-regulatioa Conclusions: The changes of cell skeletal structure and cell signal transduction might participate in the process of U937 cell apoptosis induced by Arsenic sulfide.
