摘　　要：目的 :探讨癌胚抗原重组痘病毒 (carcinoembryonicantigenrecombinantvacciniavirus,CEA -rV)治疗CEA阳性肿瘤的作用部位及其CEA基因表达的情况。方法 :采用PCR法探测CEA rV的作用部位 ,RT -PCR检测接种CEA -rV鼠腹腔MΦ 内CEA的基因表达 ,免疫组化法检测各组小鼠腹腔MΦCEA的蛋白表达。结果 :腹腔接种的CEA -rV主要感染腹腔MΦ;皮下接种的CEA -rV主要感染皮肤局部组织。CEA -rV腹腔接种小鼠 ,其腹腔MΦ有CEAmRNA表达及CEA蛋白表达。结论 :推测无论通过腹腔接种还是通过皮肤局部接种 ,GEA -rV一般多在注射局部表达。CEA -rV不仅可以进入MΦ,而且其CEA基因可以有效地表达 ,并可使MΦ 表面分子MHC -Ⅱ与B7- 2表达增强 ,然后递呈CEA给Th淋巴细胞 ,使之分泌足够的IL - 2 ,激活Tc细胞分化为CTL 。Objective: To explore the affecting location of CEA-rV on CEA positive tumor and the CEA gene expression. Methods: The affecting location of CEA-rV was probed by PCR;the CEA mRNA expression was examined by RT-PCR and the CEA protein expression was examined by immunohistochemistry. Results: Intraperitoneal CEA-rV infected mainly local M Φ;Subcutaneous CEA-rV infected mainly local tissue. CEA gene was transcripted and CEA protein was expressed only in the intraperitoneal M Φ from mice inoculated with CEA-rV. Conclusion: It could be inferred that CEA-rV infected mainly local tissue,whether inoculated by intraperitoneal or subcutaneous injection. Not only could CEA-rV infect M Φ and make M Φ express more MHC-Ⅱ and B7-2,but also enhance its CEA protein expression. Then,the CEA could be presented to Th lymphocytes by M Φ and induce adequate secretion of IL-2 to activate CTL to kill the CEA positive tumor cell.
