摘 要:目的 研究VitaminC (VC)拮抗三氧化二镍的细胞毒作用及机理。方法 采用体外细胞培养法测定在维生素C (2 5 ,5 0 ,10 0 μmol/L) )作用下 ,体外染镍肺泡巨噬细胞 (alveolarmacrophage ,AM )的存活率及活力。同时观察细胞内一氧化氮 (NO)和活性氧的产生 ,以及一氧化氮合酶 (NOS)活力的变化。结果 在体外染镍肺泡巨噬细胞中加入不同浓度的维生素C后可提高该细胞的存活率和活力 ,可减少NO和活性氧的产生 ,降低NOS和活性 ,其中以10 0 μmol/L的维生素C最为明显。 结论 镍可致细胞脂质过氧化 ,维生素C具有拮抗镍对肺泡巨噬细胞的细胞毒作用 。Objective To study the effects of V C on the injury of alveolar macrophages exposed to Ni 2O 3 in vitro.Methods The alveolar macrophages were cultured with exposure of Ni 2O 3 in vitro. Meanwhile,(25,50 and 100 μmol/L)vitmin c were added into the medium, respectively. The detections of nitric oxide (NO),active oxygen and activity of nitric oxide synthase(NOS) in alveolar macrophages were carried out. And the cell activity were determined after being exposed to Ni 2O 3 in vitro.Results The results revealed that vitamin c could decrease mortality and increase survival activity of alveolar macrophages.Active oxygen, NO and NOS activity were decreased of the same time .V C at 50 μmol /L and 100 μmol /L had the most dramatic inhibitory effect.Conclusion V C can antagonize the lipid preoxidation of alveolar macrophage induced by Ni 2O 3 ,which probably due to its antioxidant property.
