摘  要:取花生上胚轴为外植体,在农杆菌介导的基因载化过程中,在共育时加入一定量的烟草或马铃薯提取物,在共培养基和分化培养基中分别加入0 179mM的还原型谷胱甘肽(GSH)和0 147mM的VC+0 035mM的VE,进行培养。结果表明,添加15%(W/V)的烟草或马铃薯提取物,胚轴生长状况明显好于对照组,分化率较对照组提高10%~23 5%。自由基清除剂能明显减轻外植体褐化现象,促进外植体再生。GUS检测阳性率最高可达10 2%。The epicotyls from matured peanut seeds (Arachis hypogaea) were added potato or tobacco extracts and inoculated with Agrobacterium-tumefaciens.The epicotyls were then transferred to MS medium supplemented with the free radical scavenger(0179 mM GSH or 0.147 mM Vitamin C and 0.035 mM Vitamin E)and incubated at 25-26 ℃.The results showed that the rate of differentiatio increased in the range of 10% to 23.5% and epicotyls growth was much better than the controlled samples when 15%(W/V)of potato or tobacco extract was added.The brown necrosis was reduced obviously by free radical scavengers.The rate of GUS+ can reach as high as 10.2%
