8-Br-cAMP对人肺腺癌细胞P53蛋白表达和端粒酶活性的影响

摘　　要：目的 :探讨 8 Br cAMP对人肺腺癌细胞株A5 49细胞端粒酶活性和P5 3蛋白表达的影响。方法 :用终浓度分别为 2 0 μmol/L及 6 0 μmol/L的 8 Br cAMP处理人肺腺癌细胞株A5 49细胞 ,以未处理者为空白对照。采用PCR ELISA法检测端粒酶活性 ,用免疫荧光标记法测定P5 3蛋白的表达。结果 :8 Br cAMP作用后的A5 49细胞 ,端粒酶活性明显降低 ,P5 3蛋白表达明显减少。结论 :8 Br cAMP能抑制肺腺癌细胞的端粒酶活性和P5 3蛋白的表达。Aim:To investigate the influence of 8-Br-cAMP on the telomerase activity and P53 expression in the human lung adenocarcinoma cell line(A549).Methods:The human 1ung adenocarcinoma cell line(A549)was treated with 8-Br-cAMP (20 μmol/L,60 μmol/L),and the human lung adenocarcinoma cell untreated was taken as sham control. P53 expression and the telomerase activity were detected by immunohistochemisty and PCR-ELISA method in the course of differentiation of A549 cells.Results:Both P53 expression and the telomerase activity were signficantly decreased in the A549 cell line treated with 8-Br-cAMP.Conclusion:8-Br-cAMP could inhibit the telomerase activity and the P53 expression of A549 cells.
