摘 要:目的 研究人体骨骼肌失神经支配后成肌调节因子MyoD和Myf 5蛋白的表达变化 ,了解骨骼肌的再生状态。方法 2 0例不同时间人体失神经骨骼肌与 3例正常骨骼肌标本 ,HE染色和透射电镜观察肌卫星细胞形态变化 ;抗MyoD和Myf 5多克隆抗体免疫组织化学染色 (ABC法 ) ,统计阳性染色细胞核数。 结果 抗MyoD和Myf 5抗体阳性染色细胞核数在人体骨骼肌失神经 1~ 2月时的肌卫星细胞中最多 ;3个月后急剧下降 ,并维持在一低水平 ;1年后几乎无表达。结论 人体骨骼肌失神经支配后早期肌卫星细胞通过MyoD和 (或 )Myf 5途径激活。Purpose: To detect the expression of the two myogenic regulatory factors MyoD and Myf-5 protein in human skeletal muscle after peripheral nerve injury. Methods: Twenty cases denervated and 3 cases normal human skeletal muscles were involved in this experiment. Satellite cells were observed by HE staining and electric microscopy. Immunohistochemmical stain (ABC method) with polyclonal antibody against MyoD and Myf-5 protein were performed and positive nucleus were counted. Results: Denervation increased the expression of MyoD and Myf-5 protein in satellite cells. The high level of expression persisted for 1 - 2 months and low level for one year; There were rarely positive nucleus found in the cases denervated more than one year. Conclusions: Denervation of human skeletal muscle activate the satellite cells via the pathways of either MyoD of Myf-5 or both.
