碱蓬愈伤组织的诱导及继代生长

摘　　要：本实验首先在 MS基本培养基或 A号培养基 (MS+BA 0 .2 mg/ L +NAA 0 .1 mg/ L)上培养出盐地碱蓬的无菌苗 ,然后在 MS基本培养基中添加不同的激素种类及浓度配比 ,对所培养的无菌苗的下胚轴和叶进行愈伤组织的诱导与继代。结果表明 :3号培养基 (MS+2 ,4- D0 .4mg/ L+BA 0 .5mg/ L)是诱导和继代下胚轴的最适培养基 ,也是继代叶的最适培养基 ;1 2号培养基 (MS+2 ,4- D1 mg/ L+BA0 .4mg/ L)是诱导叶的最适培养基。继代数月后得到了生长旺盛、疏松、呈乳白色的愈伤组织。MS medium equipmented with different kinds and concentrations of phytohormones were used to induce and subcultrue of calli from suaeda salsa L.The culture medium No.3(MS+2,4-D 0.4mg/L+BA 0.5mg/L) was the optimal culture medium for inducing and subculturing calli from hypocotyls and for subculturing of calli from leaves,while the culture medium No.12(MS+2,4-D 1mg/L+BA 0.4mg/L) was the optimal culture medium for inducing callus from leaves.After subculture for months,quickly growing?friable?milky white calli were obtained.
