地塞米松对TNBS诱导的大鼠实验性结肠炎的治疗作用

摘　　要：目的　观察地塞米松对大鼠实验性结肠炎的治疗作用及副作用。方法　用TNBS/乙醇复制大鼠结肠炎模型 ,6h后给予地塞米松 (Dexamethasone,DX) 0 1mg/kg进行治疗 ,于处死动物前一天采血行白细胞计数并分类 ,5、7、1 0、1 4d处死动物 ,分离结肠 ,计算溃疡面积 ;取结肠粘膜测定SOD、MPO活性。结果　地塞米松能明显缩小溃疡面积 ,抑制炎症反应 ,减轻结肠重量 ,降低结肠粘膜MPO水平 ;并能升高结肠粘膜SOD的活性 ,促进溃疡愈合。在地塞米松发挥治疗作用的同时 ,可见明显的免疫抑制作用 ,表现为胸腺萎缩 ,白细胞数减少。结论　DX对TNBS/乙醇诱导的实验性结肠炎具有显著的治疗作用 ,但长期使用副作用明显 。Objective To investigate the therapeutic effects of dexmethasone(DX) on TNBS induced colitis in rats. Methods TNBS/ethanol induced colitis model was used in the experiment. Ninety rats were treated with saline or DX for 5, 7, 10 and 14d. White blood cell was counted on the day before the animals were sacrificed. Ulcerative index, SOD and MPO activity in colonic mucosa were measured. Results The remarkable effects of DX on TNBS-induced colitis were observed,with ulcerative area diminution and weight of colon descent. White blood cell population was reduced, SOD activity of colonic tissue was increased while that of MPO was decreased. In the meantime the side effects of DX, such as thymus atropy and WBC decrease, were observed during experimental period. Conclusion DX has significant effects on TNBS induced colitis in rats but also causes obvious side effects.
