摘  要:目的:探讨诱导分化剂全反维甲酸(ATRA)对肺腺癌A549细胞增殖和E-CD/catenins复合物表达的影响。方法:用细胞生长率分析不同浓度ATRA处理不同时相的A549细胞的增殖特性变化;用免疫组织化学法检测ATRA处理前后A549细胞的E-CD/catenins复合物各成分(E-CD、α-和β-catenin)的表达情况。结果:无ATRA处理组A549细胞增殖力最强,而各处理组细胞增殖能力均有较大降低;免疫组化结果显示无ATRA处理组细胞基本不表达E-CD和α-catenin,而β-catenin主要呈膜表达阳性;处理后的A549细胞E-CD、α-和β-catenin表达水平均有明显增强,并且三者的表达形式均以细胞膜表达为主。结论:ATRA可抑制肺腺癌A549细胞增殖、增强E-CD/catenins复合物表达和膜定位。Objective:To explore the effect of the differentiation-inducing agent all trans-retinoic acid(ATRA)to the cell proliferation and the expression of E-cadherin/catenin complexes in lung adenoma cell A549.Methods:After cultured by different density ATRA in different times,A549cell had been used to analyse the proliferation by the cell proliferative curve,and the expression of E-cadherin,α-catenin andβ-catenin by immunohistochemistry.Results:The proliferation was strongest in non-ATRA treated cells,but it was significantly inhibited in ATRA-treated cells.As compared with untreated A549cells,the expressions of E-cadherin,α-catenin andβ-catenin enhanced significantly in ATRA-treated cells.The expression of E-cadherin andα-catenin changed from negative to strong membranous expression and weak cytoplasmic positive.The allocation ofβ-catenin had not changed as the predominant membranous expression and weak cytoplasmic positive,but the express level enhanced.Conclusion:ATRA could enhance the expression of E-cadherin/catenin complexes and inhibit the multiplication of the lung carcino-ma cell A549.
