MMP-9与MMP-2在食管癌中的表达及其意义

摘　　要：目的 :阐明MMP 9(matrixmetalloproteinase 9)和MMP 2 (matrixmetalloproteinase 2 )在食管癌组织中的分布特点及其活性变化。方法 :采用免疫组织化学法和酶谱法 (Zymography)分别检测食管癌组织中MMP 9和MMP 2蛋白的分布及其活性。结果 :MMP 9和MMP 2在食管癌组织中的分布呈多样性 ,既存在于癌细胞中 ,也存在于成纤维细胞和组织巨噬细胞中 ;在角质细胞以及细胞间隙中也发现存在着MMP 9和MMP 2蛋白。在食管癌组织、癌旁组织和远端食管组织中存在着各种形式的MMP 9(130× 10 3-异源二聚体、92× 10 3-前酶形式和 84× 10 3-活性形式 )和MMP 2 (72× 10 3-前酶形式和6 2× 10 3-活性形式 )。 84× 10 3(活性形式 )的MMP 9以及 72× 10 3(前酶形式 )和 6 2× 10 3(活性形式 )的MMP 2在食管癌组织中的活性明显比癌旁组织和远端食管组织中的强 (P <0 0 5 )。结论 :MMP 9与MMPObjective The characteristic of distribution and activity of matrix metalloproteinase9(MMP 9) and matrix metalloproteinase2(MMP 2) was demonstrated in esophageal carcinoma tissue.Methods Immunohistochemical technique was used to reveal sites of MMP 2 and MMP 9,and the activity of MMP 9 and MMP 2 was detected by zymography in esophageal carcinoma tissue.Results The variety of distribution about MMP 2 and MMP 9 was found in esophageal carcinoma tissue.Immunohistochemical analyses indicates that MMP 2 and MMP 9 exist in esophageal carcinoma cell as well as stromal cells such as fibroblasts,macrophages,keratinocytes and cell cell.The various forms of MMP 9(dimeric form 130×10 3,pro MMP 9 92×10 3 and active MMP 9 84×10 3)and MMP 2(pro MMP 2 72×10 3 and active MMP 2 62×10 3) were found in mucosa of surgical up edge,paracancerous mucoas and cancer lesions.Activity of active MMP 9,pro MMP 2 and active MMP 2 was significantly higher in the cancer lesions than in the paracancerous mucosa and the mucosa of surgical up edge( P <0 05).Conclusions MMP 9 and MMP 2 may play important roles in pathological process of esophageal carcinoma.
