摘 要:为证实 TGF- β1 质粒 DNA能否由脂质体携带进入家兔角膜上皮细胞 ,用多聚阳离子脂质体携带重组的人TGF- β1 质粒转染体外培养的家兔角膜上皮细胞 ,在转染 12 h后 ,表达 2 d时 ,用免疫组化方法 (SABC)检测 TGF- β1质粒 DNA的表达情况。结果显示 :外源 TGF- β1 质粒 DNA在家兔角膜上皮细胞可以获得表达 ,基因转染率为 2 3.37%。提示To investigate whether the TGF β 1 plasmid DNA carried by lipofectamine could be introduced into cultured rabbit corneal epithelial cells, specific expression of the plasmid pMAMTGF β 1 in the corneal epithelial cells was detected by means of immunohistochemical method 2 days after 12 h of transfection of pMAMTGF β 1 mediated by lipofectamine. The gene transfer positive rate of the corneal epithelial cells was counted. It was found that there was foreign plasmid TGF β 1 DNA product in the corneal epithelial cells. Transfer positive rate was 23.37 %. It was suggested that efficient gene transfer into the corneal epithelial cells could be achieved by lipofectamine. Lipofectamine is an available and a promising vehicle for delivering aim gene and studying the mechanism of physiology and pathology of corneal epithelial cells.
