摘　　要：背景与目的:研究表明,加热可提高兔VX2细胞对阿霉素(adriamycin,ADM)的增敏作用,可促进ADM进入细胞内,从而提高ADM的抗癌作用。作者在兔VX2细胞行ADM热化疗研究和兔肝VX2细胞移植瘤模型建立基础上,进行ADM兔肝动脉热灌注,以评价介入性化疗与介入性热化疗对兔肝癌的抑瘤效果。方法:将VX2细胞接种于60只新西兰白兔肝右叶,建立兔肝VX2移植癌模型。随机分4组,每组15只。利用导管经肝动脉分别给各组37℃生理盐水(第1组)、37℃ADM溶液(第2组)、60℃生理盐水(第3组)、60℃ADM溶液(第4组),1周后观察各组肿瘤体积及血清AST水平,观察荷瘤兔的存活期。结果:第4组肿瘤生长率(0.53±0.21)%,与第1组(3.48±1.17)%相比有显著性差异(P<0.01),与第2组的(1.09±0.26)%、第3组的(3.32±1.28)%相比亦有显著性差异(P<0.05)。第4组存活期(87.0±2.0)天,与第1组(40.5±3.0)天相比差异有显著性(P<0.05)。第4组血清AST水平给药前后变化与其它各组相比无显著性差异(P>0.05),与对第1组相比有显著性差异(P<0.05)。结论:60℃ADM溶液热灌注可降低肿瘤生长率,延长存活期,而对肝功能的损害是可逆的。Background &Objective:It was reported that heating can enhance sensitivity of rabbit VX2cell to adriamycin and increase intracellu lar concentration of adriamycin.This study was designed to evaluate the anti-tumor effects of interventional hyperthermia and interventional chemothermotherap y on VX2carcinoma in rabbit liver.Methods :VX2carcinoma cells were surgically imp lanted into the right liver lobe of 60male New Zealand white rabbits,whic h were randomly divided into 4groups(15rabbits per group).To inject physiological saline(37℃),adriamycin(37℃),physiological saline(60℃),and adriamycin(60℃)in different groups via hepatic arte ry of the rabbits with liver cancer.One week later,to observe the volume of tumor,the serum level of aspartate transa minase(AST),and observe the survival period of VX2rabbits.Results:In group of ADM(60℃),the tumor growth rate(0.53±0.21)%was significantly lower than group2(1.09±0.26)%,group 3(3.32±1.28)%,and group 4(3.48±1.17)%(P<0.05,P<0.05,P<0.01,respectively).The survival period of adriamycin(60℃)group(50.0±2.0)d was significantly higher than the u ntreated control group(40.5±3.0)d,(P<0.05).The serum level of AST of TNP-470with lipiodol group was not higher than the other tr eated groups(P>0.05),but being significantly higher than the untreated control group after treated(P<0.05).Conclusion:Adriamycin(60℃)greatly decreases the tumour growth rate,an d prolongs the survival period.
