摘 要:通过热变性温度法(Tm法)、高效液相色谱(HPLC)法并结合全基因组测序结果对大曲高温放线菌(Thermoactinomycesdaqus)CICC 10681的(G+C)摩尔分数进行了分析,完善其遗传学特征。并对Tm法和HPLC法这两种常规测定方法进行了比较。结果显示,Tm法和HPLC法得到的CICC 10681(G+C)摩尔分数分别为47.0%(Tm)和49.3%(HPLC),与全基因组测序的结果相近,并且与已报道的高温放线菌属的(G+C)摩尔分数范围一致,符合高温放线菌低(G+C)摩尔分数的特征;与Tm法相比,HPLC法得到的(G+C)摩尔分数更准确,适合用于对微生物菌株(G+C)摩尔分数的分类学描述。The DNA(G+C) content of Thermoactinomyces daqus CICC 10681 was determined by HPLC, thermal denaturation(Tm) method combined with whole genome sequencing to know deeper on its genetic characteristics. The effect of conventional measuring method of Tm and HPLC were compared. The results showed that the DNA(G+C) mole fraction determined by Tm and HPLC was 47.0% and 49.3%, respectively. The result was similar to the whole genome sequencing result and it was also conformed to the characteristic of low(G+C) mole fraction of Thermoactinomyces sp. reported. Comparing to the Tm method, the result of HPLC method was more accurate and suitable for the(G+C) mole fraction taxonomic descriptions of microorganisms.
