摘 要:目的 研究UTX基因过表达对肾癌细胞增殖的影响.方法 通过细胞转染UTX质粒过表达UTX基因,采用实时定量PCR和western blot方法检测转染效率,运用MTT法检测其对786-O增殖的影响.结果 成功构建含有UTX基因的真核表达载体,获得UTX上调表达的786-O细胞,转染UTX质粒后肾癌细胞增殖能力显著降低,上调UTX基因表达的786-O-pCMV-UTX-HA与空白对照细胞786-O相比在24h、48h和72h细胞活力分别降低30.24%、33.88%和48.97%(P<0.01).结论 UTX基因上调可以显著抑制肾癌细胞增殖.Objectives To investigate the effects of upregulation of UTX on renal cancer cell proliferation.Methods UTX plasmids were transfected into renal cancer cell line 786-O to overexpress the UTX.The efficiancy of transfection of UTX plamid was detected by quantitive PCR and western blot.MTT assay was used to detect the effects of UTX overexpression on cell proliferation.Results Recombinant expression vector for UTX is successfully constructed and 786-O ceils highly expressing UTX are obtained.Comparing with 786-O,the cell proliferation of 786-O-pCMV-UTX-HA was significantly inhibited by 30.24%,33.88% and 48.97% after 24h,48h and 72h treatment with UTX plasmid transfection,respectively.Conclusions Up-regulation of UTX gene can inhibit cancer cell proliferation significantly.
