低氧对趋化因子FKN诱导的前列腺癌细胞转移的影响

摘  要:目的 探讨低氧对趋化因子FKN诱导的前列腺癌细胞粘附能力、侵袭能力和迁移能力的影响.方法 采用MTY法检测分别在常氧和低氧条件下经FKN诱导的PC-3细胞对细胞外基质的粘附能力,Matrigel侵袭实验检测细胞的侵袭能力,Transwell迁移实验检测细胞的迁移能力.结果 与对照组相比,FKN处理能显著提高PC-3细胞和低氧PC-3细胞的粘附、侵袭和迁移(P<0.05),相同FKN浓度时低氧条件下PC-3细胞发生粘附、侵袭和迁移的细胞数显著高于常氧条件下的细胞数(P<0.05).结论 FKN可通过增加PC-3细胞对细胞外基质的粘附作用,提高细胞的侵袭能力和迁移能力而诱导PC-3细胞发生转移,低氧可以促进FKN的作用.Objectives To investigate the effects of hypoxia on chemokines FKN induction ability of prostate cancer cell adhesion,invasion and migration ability.Methods Were determined by MTT method under the condition of oxygen and low oxygen induced by FKN PC-3 cell adhesion ability of the extracellular matrix,Matrigel invasion experiment test cell invasion ability,Transwell migration experiment test cell migration ability.Results Compared with control group,FKN treatment can significantly improve the PC-3 cells and hypoxic PC-3 cell adhesion,invasion and migration(P <0.05),the same FKN concentration under the condition of low oxygen when PC-3 cell adhesion,invasion and migration of cells was significantly higher than that under the condition of oxygen often on the number of cells(P < 0.05).Conclusions FKN can by increasing PC-3 cell adhesion effect of extracellular matrix,improve cell invasion and migration ability of inducing the PC-3 cells shift,the role of hypoxia can promote FKN.
